[g32P]GTPgAA is
a nonhydrolyzable GTP analog which has a
phenyl azide group substituted off the gamma
phosphate. [g32P]GTPgAA has
been reported to be useful for the study
G-proteins (see G-Protein
Methods, ALT
Tech Brief 1 and 2511428). With
these two structural photoaffinity isomers
of GTP, it is possible to completely map
out both the guanine base binding portion
as well as the triphosphate binding domain
of the active site of virtually any GTP binding
protein. The active site site peptides
of a protein can be easily determined using
a combination of photoaffinity labeling,
proteolytic digestion, immobilized metal
affinity chromatography (IMAC)
followed by reverse phase HPLC (see 8106359, 8439545).
